چکیده

Two kinds of prophage deletions of the Salm.onella phage P22 were isolated. One type of deletion was isolated by simultaneously selecting for loss of a host gene (pro) adjacent to the integrated prophage and for survival at the nonpermissive temperature of a temperature-inducible (tse2) lysogen. The second type of deletion was isolated by transduction to tetracycline resistance using high-frequency-transducing lysates obtained by UV-induction of an unusual tetracycline-resistant (tet ~) P22 lysogen. In the prodeletion strains, a continuous block of genes beginning beyond the left end of the prophage and including all or part of proA is deleted. The c-linked immunity region (immC) is deleted in all the pro-deletion strains. In contrast, in the tet R deletions a continuous block of genes beginning at the right (proC) end of the prophage is deleted. The ran-linked immunity region (immI) is deleted in all of the tet R deletions. These two sets of prophage deletions confirm the established prophage gene order and have been used to compile a comprehensive deletion map of the P22 prophage which includes many previously unmapped genes. The bipartite nature of immunity in P22 is confirmed by the observation that both prodeletions, which laek immC but retain imrnI, and tet~ deletions, which lack immI but retain immC, are each sensitive to superinfection by homoimmune nonvirulent P22 phage.

Regional specificity of illegitimate recombination by the translocatable ampicillin-resistance element Tn1 in the genome of phage P22.

Insertions of the translocatable ampicillin-resistance element Tn1 were selected in the genome of the temperate Salmonella phage P22 by growing the phage on hosts carrying the resistance plasmid RP4. Insertions of Tn1 into phage P22 are rare (10(-10) per phage) and nonrandomly distributed in the P22 genome. They are found mainly in the vicinity of the P22 ant gene. Insertions within the ant gen...

Genetics of bacteriophage P22. II. Gene order and gene function.

Although large numbers of temperate phages are known to exist in nature, only a small number of these have been examined in much detail. Foremost among these is the coliphage ~, which has been studied in great depth over the past two decades. However, it is already quite clear that phage ~ represents only one alternative among many with respect to any of its interesting properties. Most other t...

Dual control of lysogeny by bacteriophage P22: an antirepressor locus and its controlling elements.

Two distantly linked clusters of genes on the Salmonella typhimurium phage P22 chromosome are involved in the control of lysogeny and superinfection immunity. One cluster consists of genes cl, ~2, and ~3, which are primarily responsible for the establishment and maintenance of lysogeny. It has been proposed that the second cluster consists of three loci which are responsible for the synthesis a...

Superinfection exclusion by P22 prophage in lysogens of Salmonella typhimurium. II. Genetic evidence for two exclusion systems.

Mutants of the Salmonella phage P22 which, as prophages, do not prevent the growth of superinfecting virulent P22 phage were isolated. These mutants, called sieA-, retain some ability to exclude the heteroimmune phages L and MG178. Prophages carrying mutations at another locus, sieB, lose entirely the ability to exclude the heteroimmune phages. The sicmutants of Rao (1968) were shown to be sieA...
